By employing myriocin, an inhibitor of serine palmitoyltransferase, which catalyzes the initial step in sphingolipid biosynthesis, the levels of sphingomyelin, sphingosine and glycosphingolipids are all reduced [28]. Sphingomyelin is the major product of the sphingolipid biosynthetic pathway, and spectrophotometric analysis of myriocin-treated wt fibroblasts (with or without U18666Atreatment) and NPC1-mutant fibroblasts confirmed that myriocin was able to decrease the amount of sphingomyelin in these cells by at least 40 (Figure 3A). Of note, in a similar experimental setting filipin staining was JI-101 web demonstrated to be diminished by prolonged myriocin treatment [22].
